Random Primer DNA Labeling Kit(TAKARA)

[α-32P], [3H]dCTP DNA를 표식하여 hybridization DNA 프로브를 제작하는 키트이다. Feinberg Vogelstein의 방법을 개량하여 간단한 조작으로 높은 비방사활성의 DNA 프로브를 제작할 수 있다.

Random Primer DNA Labeling Kit Ver.2 3’→ 5’exonuclease 활성을 제거한 Exo-free Klenow Fragment Random Primer (9 mer)를 조합하여 표식하므로써 1×109dpm/㎍의 높은 비방사활성 프로브를 단시간 (10분 이내)에 제작할 수 있다.

 

시약

Random Primer (9mer)

10x Buffer

dNTP Mixture

Exo-free Klenow Fragment (2U/ul)

Control DNA (λ-Hind III Fragment 25 ng/)

 

보관: -20℃

 

1) Combine the follow reagents in a microcentrifuge tube and bring the total volume to 14ul with distilled sterilized water or TE buffer.

Template DNA 10ng ~ 1ng

Random primer 2ul

sterile distilled water or TE buffer n ul

total 14ul

 

2) Heat at 95 for 3minutes and then cool on ice for 5minutes.

 

3) Add 2.5ul of 10xbo, 2.5ul of the dNTP mixture, and 5ul of labeled dCTP. (1.85MBq, 50u Ci)

 

4) Add 1ul of Exo-free Klenow fragment and incubate at 37 for 10minutes.

 

5) Inactivate the enzyme by heating the mixture at 65 for 5minutes of adding EDTA in a final concentration of 30mM. > 0.2M EDTA > 3.75ul 사용

 

6) Heat at 95 for 3minutes and then cool on ice.

 

7) Use an appropriate volume as hybridization probe. (When necessary, unincorporated dCTP is removed by gel filtration of by ethanol precipitation.)
