- 2x Rapid Ligation Bo, T4 DNA ligase : 5ul
- pGEM-T Easy vector(50ng) : 1ul
- insert DNA : 3ul (양에 따라서 16ng/ul 일때 3ul 넣음)
- T4 DNA Ligase(3 Weiss units/ul) 1ul
- Deionized water to a final volume of : 10ul

2. E.coli transformation
- competent cell(BIOFACT cat.no.SOT41-K020) ice에 녹일 것, ligate도 같이 ice에 놓을 것, incubate 미리 켜놓을 것 42℃
- ligate mixing (pepetting하지 않을 것)
- ice, 10~30min
- heat, 42℃, 1m30s
- ice, 2min
- LB, 900ul 첨가 37℃, shaking incubate, 1hr
- LB/100Amp 배지에 IPTG/X-Gal(47ul(7/40) 도말
- 100ul 도말
37℃ O/N
3. 오전에 plate 꺼낸 후, 오후에 colony 1개 각3개씩 LB/100Amp 액체 배지 8hr이상 배양

4. colony prep 후 sequence 맡김
